IGF-1 LR3 is a modified recombinant analog of insulin-like growth factor 1, carrying an arginine substitution at position 3 and a 13-residue N-terminal extension. Both modifications serve the same purpose: they sharply reduce binding to the IGF binding proteins (IGFBPs) that normally sequester circulating IGF-1. Since roughly 98% of native IGF-1 in circulation is bound to IGFBPs at any moment, an analog that escapes them behaves very differently as a laboratory reagent — which is why LR3 became a standard supplement in mammalian cell-culture media.
Characterised as an IGF-1 receptor agonist with markedly reduced IGFBP affinity. Published work has compared the differential effects of IGF-1 and gonadotropins on proliferative activity in distinct granulosa cell subgroups, and examined interactions between follicle-stimulating hormone and growth factors in DNA synthesis in bovine granulosa cells — cell-culture studies of the receptor system this analog engages. Not FDA-approved for human use; IGF-1 and its analogs appear on the WADA Prohibited List.
