Semax is a synthetic heptapeptide developed at the Institute of Molecular Genetics of the Russian Academy of Sciences in the 1980s. It is built from ACTH(4–10) — a fragment of adrenocorticotropic hormone already known to be behaviourally active but rapidly degraded — with a Pro-Gly-Pro tail added at the C-terminus specifically to slow enzymatic breakdown. Critically, the fragment was chosen because it lacks the corticotropic activity of full-length ACTH: the design intent was to keep the neuroactive portion and discard the hormonal one. It is listed in the Russian pharmacopoeia but has never been approved by the FDA or EMA.
The characterised biology sits in two areas. First, ACTH(4–10)-derived peptides interact with melanocortin receptors and with BDNF expression in rodent brain tissue. Second — and less widely appreciated — the N-terminal acetylation state of Semax substantially changes its coordination of copper(II) and zinc(II) ions, which a 2016 study in J Inorg Biochem tied to its biological properties. A 2021 rodent study examined behavioural and neurochemical measures following early-life fluvoxamine exposure. This is animal and in-vitro work; Semax is not FDA-approved for any use.
