Origin

Retatrutide (development code LY3437943) is a synthetic single-chain peptide engineered to engage three receptors at once — GIP, GLP-1 and glucagon. It is the third generation of a design lineage that began with single GLP-1 receptor agonists and moved through dual GIP/GLP-1 agonists: each step adds a receptor to the same peptide backbone. The engineering problem is balancing potency across three receptors in one molecule.

What has been characterised

Characterised as a triple agonist at the glucose-dependent insulinotropic polypeptide receptor (GIPR), the glucagon-like peptide-1 receptor (GLP-1R) and the glucagon receptor — all three class-B G-protein-coupled receptors. The receptor-level literature cited below covers the structural biology of these targets: the cryo-EM structure of the activated GLP-1 receptor in complex with a G protein, and a review of GIP and its receptor.
