The incretin peptides are an engineering lineage rather than a set of discoveries: single GLP-1 receptor agonists came first, then dual GIP/GLP-1 agonists built on the same backbone, then triple agonists adding glucagon receptor activity — each generation adding a receptor to one peptide chain, with the design problem being how to balance potency across all of them at once. The amylin analogues engage a different receptor family again. Alongside them sit two small molecules that are not peptides at all: 5-Amino-1MQ, an NNMT enzyme inhibitor, and SLU-PP-332, a synthetic ERR agonist studied in mitochondrial biogenesis work.
