Background & literature
Origin
Melanotan I is the linear alpha-MSH analog [Nle4, D-Phe7]-α-MSH, developed at the University of Arizona by Victor Hruby and Mac Hadley from the 1980s. The two substitutions — norleucine at position 4 and D-phenylalanine at position 7 — were made to resist enzymatic degradation and increase receptor potency relative to native α-MSH. Under the name afamelanotide it went through pharmaceutical development and was approved in the EU and later the US for a rare photosensitivity disorder.
What has been characterised
Characterised as an agonist at the melanocortin-1 receptor (MC1R), the receptor on melanocytes whose activation drives eumelanin synthesis via the cAMP pathway. Its selectivity for MC1R over the other melanocortin receptors is the pharmacological feature separating it from melanotan II. A 2026 review in Dermatologie covers hormones and skin pigmentation and the clinical relevance of this receptor system.
