Background & literature
Origin
Melanotan II is the cyclic lactam analog of alpha-MSH, developed alongside melanotan I at the University of Arizona. Cyclisation constrains the peptide’s conformation, which increased potency but cost the receptor selectivity that melanotan I has — MT-II is a broad melanocortin agonist. That lack of selectivity is precisely what produced the off-target observations during early testing that led, indirectly, to the development of bremelanotide.
What has been characterised
Characterised as a non-selective melanocortin receptor agonist with activity at MC1R, MC3R, MC4R and MC5R. Published rodent work has examined MT-II microinjected into the nucleus accumbens, and studied it in PACAP-deficient mice, in both cases probing central melanocortin receptor signalling — animal studies of central melanocortin signalling. Not FDA-approved for any use, and it has been the subject of FDA and international regulatory warnings.
