Sermorelin is the synthetic 1–29 amino acid fragment of human growth-hormone-releasing hormone — the shortest fragment of the 44-residue parent that retains full receptor activity, which is why it became the reference GHRH analog. It entered clinical research in the 1980s and was studied through the early 1990s, including in paediatric endocrinology, before being withdrawn from the US market in 2008 for commercial rather than safety reasons.
Characterised as an agonist at the GHRH receptor on anterior pituitary somatotrophs, acting on the same receptor as endogenous GHRH but as a truncated 29-residue peptide. A 1990 study examined continuous subcutaneous GHRH(1–29)NH2 over a year in short, slowly growing children — historical clinical work on the parent molecule, and among the reasons sermorelin is the best-characterised member of this class. Not currently FDA-approved; GHRH analogs are on the WADA Prohibited List.
