Background & literature

Origin

Kisspeptin has one of the better naming stories in biochemistry. The parent gene KISS1 was identified in 1996 as a metastasis-suppressor in Hershey, Pennsylvania, and the researchers named the product “kisspeptin” after Hershey’s Kisses. Its role as the upstream regulator of GnRH release was discovered separately and later, when loss-of-function mutations in its receptor GPR54 were found in patients with hypogonadotropic hypogonadism. Kisspeptin-10 is the C-terminal decapeptide fragment that retains full receptor activity.

What has been characterised

Characterised as an agonist at KISS1R (GPR54), a G-protein-coupled receptor, positioned upstream of GnRH neurons in the hypothalamic-pituitary-gonadal axis. Published work has examined kisspeptin-10 binding to Gpr54 in osteoclasts with respect to Dusp18-mediated dephosphorylation of Src, and studied KISS1/KISS1R expression in prostate tissue.
